The bootloader is completely corrupted, or the USB drive is unreadable. Try switching to a smaller, older USB 2.0 drive, or change the name of the firmware file to match the exact generic naming convention required by the board's bootloader code.
Solder fine enameled copper wires from those test pads to your programmer's corresponding In-System Programming (ISP) header. Connect a common Ground (GND) wire.
